The spread of cancer cells from their organ of origin to distant tissues is called metastasis. Cancer metastasis is the main cause of death from cancer, and in many cases is difficult to detect or treat. The process by which tumour cells become metastatic is complex and involves many stages, including detachment of cells from the main tumour mass, degradation of the surrounding extra-cellular matrix, invasion into nearby blood vessels, travel and survival through the circulatory system, attachment to a vessel wall, extra-vasation, degradation of the extra-cellular matrix into a distant tissue/organ, and the development of a novel blood supply. In order to accomplish this process, the cells acquire characteristics which are important for each stage. Recently, a class of genes known as metastasis suppressors' has been the subject of intense investigation. For some metastasis suppressor genes, there is strong evidence from both in vitro and in vivo studies to demonstrate key roles in the metastatic process, for others data is much more limited, and their importance uncertain. In this book, chapters are devoted to providing up-to-date summaries of our understanding of individual metastasis suppressor genes. Each is written by a leading authority in the study of that gene. Topics covered include discussions on how each metastasis suppressor was discovered, the mechanisms underlying their loss of expression in tumours and tumour cell lines, their proposed molecular functions, and the consequences to a tumour cell of the loss of this function. This compilation aims to provide, in a single volume, comprehensive information that will be valuable to all scientists working in cancer research, to students needing to understand molecular events that regulate tumour progression and the acquisition of metastasis, and to clinicians who might wish to know more of the roles of potentially new markers for cancer diagnosis and prognosis.

Par-4 is a naturally occurring tumor suppressor. Studies have indicated that overexpression of Par-4 selectively induces apoptosis in cancer cells while leaving normal, health, cells unaffected. Mechanisms contributing to this cancer-selective action of Par-4 have been associated with PKA activation of intracellular Par-4 in cancer cells or GRP78 expression primarily on the surface of cancer cells. On the other hand, endogenous Par-4 sensitizes cells to the action of a broad range of apoptotic inducers acting via the extrinsic and intrinsic pathways. A number of binding partners of Par-4 have been identified and shown to regulate Par-4 function in cancer and other diseases, such as Alzheimer’s and major depression. Recent studies have recognized a number of natural products, dietary supplements, synthetic molecules and FDA-approved drugs that induce the secretion of Par-4 protein to cause apoptosis in primary or metastatic tumors, one of which is in clinical trials. Metastasis-suppressor genes suppress the growth of metastases without affecting tumor growth. We have been studying the role of inactivation of one such metastasis suppressor gene, Map Kinase Kinase 4 (MKK4) in the process of metastatic colonization. Work proposed in this application was aimed at extending our ongoing studies in the AT6.1 model system into established transgenic models of prostate cancer and the use of transgenic approaches to test further MKK4's metastasis suppressor activity. Over the finding period of this grant we worked to test our hypotheses appropriately. During the course of these studies we found a need for changing our experimental design. To address these we established new collaborations and re-examined the potential role of MKK4 in tumorigenesis in both the TRAMP model and human cancers. In pursuing these studies we identified an unanticipated role MKK4 in the early growth of primary tumors and disseminated cells at metastatic sites.
