When the plasma clotted, it formed a solid surface into which the heart cells explanted. Left unattended, these cells died within several days. By regularly feeding the grow ing cells with aqueous extracts of whole chick embryos and periodically sub dividing them, Carrel was able to maintain the cells for extended periods. From these initial experiments, cell culture has expanded into an important component of biological research and commercial production. Animal cell culture is an important tool for the study of complex biologi cal systems. In vivo it is often impossible to target a treatment to one specific cell type or to adequately control the environment. For example, it is impos sible to selectively depolarize endothelial cells in vivo to determine the role of transmembrane potential in the transduction of fluid mechanical forces generated by blood flow to a biochemical response. Depolarizing all of the cells within the vascular system by injecting potassium chloride into the animal's bloodstream clearly is not an option, as it will lead to cardiac arrest.

1 Kevin Moses It is now 25 years since the study of the development of the compound eye in Drosophila really began with a classic paper (Ready et al. 1976). In 1864, August Weismann published a monograph on the development of Diptera and included some beautiful drawings of the developing imaginal discs (Weismann 1864). One of these is the first description of the third instar eye disc in which Weismann drew a vertical line separating a posterior domain that included a regular pattern of clustered cells from an anterior domain without such a pattern. Weismann suggested that these clusters were the precursors of the adult ommatidia and that the line marks the anterior edge of the eye. In his first suggestion he was absolutely correct - in his second he was wrong. The vertical line shown was not the anterior edge of the eye, but the anterior edge of a moving wave of patterning and cell type specification that 112 years later (1976) Ready, Hansen and Benzer would name the "morphogenetic furrow". While it is too late to hear from August Weismann, it is a particular pleasure to be able to include a chapter in this Volume from the first author of that 1976 paper: Don Ready! Mechanobiology is now a vigorous branch of biomechanics and biorheology which is mainly concerned with the study of the influence of mechanical forces on cells and tissues and their clinical or therapeutical applications. As we are now at the age of proteomics and genomics and of cell micromechanical approaches, using methods like laser tweezers or confocal microscopy, mechanobiology brings new challenges. With these new researches, mechanobiology is the promise of new diagnostic and therapeutic approaches. T to the system under scrutiny and that stresses are implicated in tissue physiology (for example by the production of the extracellular matrix), secretions (i.e. production of NO and prostaglandins by endothelial cells), or for the induction of specific functions via intercellular communication; hence the interest from pharmacology in studies on new molecules. The partition of fluid between the vascular and interstitial compartments is regulated by forces (hydrostatic and oncotic) operating across the microvascular walls and the surface areas of permeable structures comprising the endothelial barrier to fluid and solute exchange, as well as within the extracellular matrix and lymphatics. In addition to its role in the regulation of vascular volume, transcapillary fluid filtration also allows for continuous turnover of water bathing tissue cells, providing the medium for diffusional flux of oxygen and nutrients required for cellular metabolism and removal of metabolic byproducts. Transendothelial volume flow has also been shown to influence vascular smooth muscle tone in arterioles, hydraulic conductivity in capillaries, and neutrophil transmigration across postcapillary venules, while the flow of this filtrate through the interstitial spaces functions to modify the activities of parenchymal, resident tissue, and metastasizing tumor cells. Likewise, the flow of lymph, which is driven by capillary filtration, is important for the transport of immune and tumor cells, antigen delivery to lymph nodes, and for return of filtered fluid and extravasated proteins to the blood. Given this background, the aims of this treatise are to summarize our current understanding of the factors involved in the regulation of transcapillary fluid movement, how fluid movements across the endothelial barrier and through the interstitium and lymphatic vessels influence cell function and behavior, and the pathophysiology of edema formation.
