Book Description
Gliomas are fatal diseases, but also represent good models for tumor research with the aim to eventually discover new and appropriate therapeutics against this disease. Glioma experimental research models are of help to investigate tumorigenesis (tumor stem cell theory versus "classical" opinions), tumor angiogenesis (since they are highly vascularized) and tumor invasion (since they grow without limits). In addition, they have a very special microenvironment (the brain) and limited tumor stroma cells (mainly microglia and endothelial cells). This book addresses the molecular mechanisms of the various tumor stages, describes the interaction with the tumor microenvironment and furthermore depicts experimental models for Glioma research and future therapeutic concepts. The book is composed and written for Scientists and Medical Doctors in Oncology, Neurosciences and Molecular Biology.

Book Description
This volume presents the most current reviews on how cancer stem cells (CSCs) hypothesis dictates that the continued proliferation of a tumor is dependent on a sub-population of self-renewing and asymmetrically dividing neoplastic stem cells that supply a largely differentiated tumor. This volume provides a comprehensive overview of the characteristics of CSCs, their role in central nervous system (CNS) tumors, and the recent CSC-specific treatment modalities being used. The emerging focus on CSCs in brain tumors represents a paradigm shift in our understanding of the pathogenesis of these neoplasms. Importantly, the realization that a distinct sub-population of cells contributes disproportionately to the growth and sustenance of central nervous system tumors has important implications for the treatment of such tumors. To treat CNS tumors, there is now a growing need to treat CSCs to achieve adequate tumor control.

Book Description
Gliomas, developing in the brain from the transformed glial cells, are a very special kind of tumor, extremely refractory to conventional treatments. Therefore, for the development of new antitumor strategies, a better understanding of molecular mechanisms responsible for their biology, growth and invasion is still needed. This book is a reference on cellular signaling processes regulating gliomas physiology and invasiveness. The work is focused on the mechanism of nucleotide receptor activation by exogenous nucleotides and formation of complex signaling cascades induced by growth factors, cytokines and cannabinoids. The second edition of the book enriched in new chapters provides a framework explaining how signal transduction elements may modulate numerous genetic and epigenetic alterations, describes the role of local microenvironment in cellular growth, progression and invasion and, in the light of extensive new results, presents perspectives concerning potential targets for gliomas therapy.

Book Description
A dramatic increase in knowledge regarding the molecular biology of gliomas has been established over the past few years, and this has led to the development of novel therapeutic strategies for these patients. This book describes some improvements in the surgical management of gliomas, including segmentation of brain MRI images using 4D MRI volumes to help with the diagnosis and monitoring of patients. Another novel topic reviewed involves the applications of photosensitizers and their efficacy in the generation of anti-tumor responses in photodynamic therapy. A review of the application of nanoparticles and their ability to deliver drugs to the tumor site with a reduction in systemic toxicity is another developing therapy discussed. The book also describes novel approaches involving the development of the use of microRNAs, which are non-coding RNAs that can be used as tumor suppressors that potentially can be developed to control the growth of gliomas. The book examines a large number of molecular interactions of signals in gliomas, which should lead to biomarkers of potential importance that could be manipulated in the development of clinical trials. Molecular networks need to be better understood for the development of therapeutic strategies. Finally, the book reviews immunotherapeutic strategies potentially useful in treating brain tumors that involve either poxviruses engineered to secrete IL-15- or IL-2-secreting fibroblasts transfected with tumor DNA. The stimulation of the immune system to selectively attack malignant cells should lead to the prolongation of survival of brain tumor patients without a decline in cognitive functions or other side effects. It is hoped that this new information will lead to improved and efficacious treatment strategies for these challenging tumors.
