The SourceBook series was inspired by our difficulty in locating a suitable replacement for a depleted reagent in the midst of an urgent research project. The architectures (three-dimensional shapes) of peptides determine their respective biological functions. Therefore, the correct alignment of functionalities in a structure by constraining the flexibility is a key process in evolution as well as in medicinal chemistry in order to increase binding affinity and selectivity. The rigidification of a peptide chain can have local effects (incorporation of the amino acid proline) or it can globally restrain flexibility (macrocyclization). Furthermore, the combination of both strategies has given rise to complex antibiotics with highly optimized modes of action. This work approaches these principles in three topics and for different purposes. The first chapter presents a novel scanning strategy which utilizes synthetic local rigidifications for the evaluation of Neuropeptide Y structure and receptor binding patterns. The fundamental process of macrocyclization is topic of the second chapter. For iminopeptides, ring-chain equilibria can be established and controlled, thereby allowing for the thermodynamic analysis of the ring closure. This leads to the identification of structural determinants that influence the inclination of a peptide chain to close the ring. In the third chapter, a sugarbased synthetic pathway leading to highly functionalized thiazole dipeptides is described. This strategy led to the synthesis of core motivs of complex thiopeptide antibiotics, as well as to diastereomers and homologs thereof.

This thesis reports the calculation of neutrino production for the T2K experiment; the most precise a priori estimate of neutrino production that has been achieved for any accelerator-based neutrino oscillation experiment to date. The production of intense neutrino beams at accelerator facilities requires exceptional understanding of chains of particle interactions initiated within extended targets. In this thesis, the calculation of neutrino production for T2K has been improved by using measurements of particle production from a T2K replica target, taken by the NA61/SHINE experiment. This enabled the reduction of the neutrino production uncertainty to the level of 5%, which will have a significant impact on neutrino oscillation and interaction measurements by T2K in the coming years. In addition to presenting the revised flux calculation methodology in an accessible format, this thesis also reports a joint T2K measurement of muon neutrino and antineutrino disappearance, and the accompanying electron neutrino and antineutrino appearance, with the updated beam constraint.

FUEGER Among the many processes in Physiology few appear mo~e inviting to be studied by tracers and external imaging than the variety of the routes of migratory (blood) cells in health and disease. Much emphasis has been placed lately on the methods of labelling of the white blood cells. It is obviously quite important and necessary to refine the methods of leucocytic labelling, particularly to search for ways to label selectively a specific group of white blood cells, but there is also the need to review and keep abreast with the developing knowledge of the white blood cells themselves, especially their behaviour under pathological conditions, as seen by histology and scintigraphy, their biological properties, their immunological characteristics and the mechanisms of the control of leucocytic functions. Similarly, it appears desirable to analyze animal models of inflammation as well as to review the dosimetry and the biodistribution of labelled white blood cells in humans. This book is the result of a cooperative effort to review certain highlights of the physiology of leucocytes, labelled and unlabelled, as a corollary to the effort concerning the labelling of white blood cells.
