This volume will provide a contemporary account of advances in chemical carcinogenesis. It will promote the view that it is chemical alteration of the DNA that is a route cause of many cancers. The multi-stage model of chemical carcinogenesis, exposure to major classes of human carcinogens and their mode-of-action will be a focal point. The balance between metabolic activation to form biological reactive intermediates and their detoxification, ensuing DNA-lesions and their repair will be profiled. It will describe the chemical changes that occur in DNA that result from endogenous insults including epigenetic changes that lead to gene silencing. The sixth annual research conference of the American Institute for Cancer Research was held August 31 and September 1, 1995, at the Loews L'Enfant Plaza Hotel in Washing ton, DC. In view of the promising leads in the diet/nutrition and cancer research field, the conference was devoted to "Dietary Phytochemicals in Cancer Prevention and Treatment. " The number of sessions was increased over that in previous conferences in order to accommodate the topics of interest. The conference overview, entitled "Plants and Cancer: Food, Fiber, and Phytochemicals," provided a framework for the following sessions. In addition, the attendees were reminded that for several decades epidemiologists have noted a lower risk of lung, esophageal, stomach, and colon cancer in populations consuming diets high in fruits and vegetables. However, isolation and ingestion of individual protective factors are not the preferred action since the complexity of the food and the matrix in which nutritional factors are embedded are important. The individual sessions then provided more insight as to why eating fruits and vegetables is associated with a lower risk of cancer. The first of these sessions was on "Isothiocyanates" that induce both the Phase I and Phase II enzymes that increase detoxification and conjugation reactions, thus causing more rapid removal of any xenobiotic or carcinogen. Thus, less carcinogen is available for interaction with DNA or other critical cellular macromolecules.

The Eighth Annual Research Conference of the American Institute for Cancer Research, held in Washington, D.C., September 3-4, 1998, was on the subject "Colon Cancer Prevention: Dietary Modulation of Cellular and Molecular Mechanisms," with participants representing various disciplines interested in this area. One of the speak ers provided an appropriate quote from 17th century physician Thomas Adams: "Pre vention is better than healing because it saves the labor of being sick," which aptly describes the need for the prevention of cancer. An overview of normal and abnormal colonic development emphasized that although the typical human colon undergoes 1013 cell divisions by age 60, with the asso ciated possibilities for error, relatively few colon tumors develop. Since dietary modu lation leads to extremely small changes in colonic cells over a long period, animal models are useful to time, observe, and delineate the events associated with colon cancer. In the development colon cancer, the inactivation of the adenomatous polyposis coli (Apc) gene is one of the earliest known events. Normally Apc downregulates the cellular protein beta-catenin, but this is lost during cancer development. Beta-catenin may itself be an oncogene; it has a short half-life, but it is stabilized by binding to is more prevalent in the cell nucleus, the gene shuttles caherin.
