The development of PCR, which enables extremely small amounts of DNA to be amplified, led to the rapid development of a multiplicity of a- lytical procedures that permit use of this new resource for the analysis of genetic variation and for the detection of disease-causing mutations. The advent of capillary electrophoresis (CE), with its power to separate and a- lyze very small amounts of DNA, has also stimulated researchers to develop analytical procedures for the CE format. The Canadian Forces require rapid, sensitive systems for identification of bacterial & viral biological warfare (BW) agents in environmental matrices such as air and water. The system must detect BW agents under battlefield conditions at extremely low concentrations with no false alarms. Together, capillary zone electrophoresis with laser induced fluorescent detection (CE-LIF) of gene probe fragments offers the potential for developing a sensitive BW agent identification capability. Gene probes can be used to target signature sequences of BW agents for identification, and CE-LIF detection is an attractive technology for detection of nucleic acid fragments because of its ultra-low detection limits and relatively simple instrument design. This paper describes experiments using a probe for Bacillus globigii, an anthrax simulant, to demonstrate the capabilities of CE-LIF technology with regard to identifying target DNA, the detection limits achieved, ease of use, and speed of detection compared to other methods.

A method is provided for sizing DNA fragments using high speed detection systems, such as flow cytometry to determine unique characteristics of DNA pieces from a sample. In one characterization the DNA piece is fragmented at preselected sites to produce a plurality of DNA fragments. The DNA piece or the resulting DNA fragments are treated with a dye effective to stain stoichiometrically the DNA piece or the DNA fragments. The fluorescence from the dye in the stained fragments is then examined to generate an output functionally related to the number of nucleotides in each one of the DNA fragments. In one embodiment, the intensity of the fluorescence emissions from each fragment is linearly related to the fragment length. The distribution of DNA fragment sizes forms a characterization of the DNA piece for use in forensic and research applications.
