In-situ chemical oxidation (ISCO) is based on the delivery of oxidants to the subsurface where the oxidant is consumed by the contaminants of concern as well as by natural organic matter and other reduced species within the soil. The amount of oxidant consumed by the soil is known as the natural oxidant demand (NOD). During ISCO, the oxidant can change the physical and chemical structure of the soil, altering the microbial environment, and killing microorganisms. The following study was performed to assess the effects of chemical oxidation on NOD removal and microbial activity as expressed through the biodegradation of toluene. The oxidants evaluated as part of this study included sodium percarbonate, hydrogen peroxide, sodium persulfate, and potassium permanganate. Each of the oxidants was evaluated at low, medium and high dosages. A permanganate chemical oxygen demand (PCOD) test was used to determine the pre- and post-treatment NOD of the soil. Following chemical oxidation, each soil sample was added to a biometer flask, seeded with the microbe Pseudomonas putida F1, and then an aliquot of aqueous phase toluene was added to each biometer flask. The CO2 production from each sample was monitored with time to evaluate the oxidation effects on microbial activity. The oxidants potassium permanganate, sodium persulfate, sodium percarbonate, and hydrogen peroxide all removed some permanganate NOD (PNOD) with permanganate expressing the largest removal of PNOD. Each of the oxidants affected microbial CO2 production with permanganate, hydrogen peroxide, and sodium persulfate oxidation resulting in increased in overall CO2 production. Sodium percarbonate oxidation resulted in decreasing CO2 production. The ISCO process adversely affected microbial activity only when percarbonate was used.
